One of the main causes of no
n-specific background staining is the binding of antibodies to Fcγ receptors (FcγR), making many experiments involving cells expressing FcγRs difficult to interpret.
Fc Silent™ antibodies contain engineered Fc domains with abolished FcγR binding. Fc Silent™ does not affect other properties of the antibody, including their compatibility with secondary antibodies.
Fc Silent™…and that’s it!
As an example, we show how murine macrophages are labelled almost as strongly by a rat IgG2b isotype control as by an anti-F4/80 (Figure 1). Switching to Fc Silent™ antibodies results in reliable data with background staining indistinguishable from a secondary-antibody-only control.
This also shows that Fc Silent™ antibodies are compatible with standard secondary antibodies.

Compared to Fc receptor blocking reagents, Fc Silent™ antibodies do not require additional steps in your experiment and are more widely compatible with secondary antibodies than F(ab’)2 and Fab fragments.
