Recombinant monoclonal antibody to IL-2. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma LNKB-2.

Specificity: This antibody binds human IL-2 at the 59-72 site of the IL-2 sequence.
Application Notes: This antibody was quantified by ELISA and used for latex-agglutination analysis of human IL-2. It was measured to bind human IL-2 with high affinity (Ka ≈ 3 × 108 M-1) (Lunev et al., 1990; PMID: 1718463). This antibody has been used successfully for WB and FACS (Stratieva-Taneeva et al., 1993; PMID: 8359822). The crystal structure characterization of the Fab version of this antibody was performed (Fokin et al., 2000; PMID: 11040993). Further crystallization of the Fab version of this antibody with an antigenic peptide revealed that the antibody-antigen complexation involves a significant rearrangement of the epitope-containing region of the IL-2 with retention of the α-helical character of the epitope fragment (Afonin et al., 2001; PMID: 11468348).

Western Blot using anti-IL-2 antibody LNKB-2 (Ab00232) Untreated human peripheral blood monocyte (PBM) (A), PMA/lonomycin-treated PBM (B), human spleen (C), human thymus (D), Jurkat cell (E) and Molt4 cell (F) lysate samples (35µg protein in RIPA buffer) were resolved on a 15% SDS PAGE gel and blots probed with the rabbit IgG-chimeric version of LNKB-2 (Ab00232-23.0) at 1 µg/ml (A, B) or 0.1 µg/ml (C-F) before detection using an anti-rabbit secondary antibody. Protein was detected by chemiluminescence. The expected running size for IL-2 is 17.6kDa. Ab00232-23.0 successfully detected Il-2 in multiple human tissue / cell samples.
