Recombinant monoclonal antibody to CD6. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma OX-126.

Immunogen: This antibody was raised by immunizing BALB/c mice with a fusion protein consisting of human CD6 domain 3 and rat CD4 domains 3 and 4 (CD6d3CD4d3 + 4) (corrected, see: (Hassan et al., 2019; pmid: 30890626)).
Specificity: This antibody is specific for the domain 2 of human CD6. Binding depends on a SNP ( R, amino acid 225).
Application Notes: This antibody is noncompetitive with OX125 and does not block CD6/CD166 interactions (Santos et al., 2019; pmid: 31162836).

Immunofluorescence staining of human peripheral blood leukocytes using anti-CD6 (Ab00539) OX-126 Immunofluorescence analysis of paraformaldehyde fixed human peripheral blood leukocytes stained with the chimeric mouse IgG version of OX-126 (Ab00539-1.1) at 10 µg/ml followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00539-1.1, DAPI, merged channels and an isotype control. The isotype control was stained with anti-unknown antibody (Ab00178-1.1) followed by Alexa Fluor® 488 secondary antibody.
Flow cytometry using the Anti-CD6 antibody OX-126 (Ab00539). Paraformaldehyde fixed human peripheral blood monocytes were stained with anti-unknown specificity antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG1 version of OX-126 (Ab00539-1.1, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-mouse IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
