Recombinant monoclonal antibody to NCAM-L1. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the phage display antibody H2 scFv.

Immunogen: single-chain variable fragment antibodies against mouse L1 from a human synthetic phage display library.
Specificity: This scFv binds specifically to the fibronectin type III repeats 1–2 of mouse L1 with a KD of around 2μM (determined by ELISA). It does not cross-react with human L1 in ELISAs, Western blots or immunofluorescence staining of cultured neurons. L1 plays important roles in cell adhesion, neuronal migration, neurite outgrowth, fasciculation and pathfinding, neuronal survival, and synaptic plasticity. L1 is not recongised under reducing conditions. L1 is a membrane glycoprotein of the immunoglobulin superfamily. Its interacts with adhesion molecules and proteins of the ECM. L1 also is inolved in the migration of cerebellar granule cells, enhancing neuronal survival and memory.
Application Notes: This scFv can stimulate neurite outgrowth from cerebellar dorsal, rootganglion and motor neurons and also be used to enhance survival of cerevellar neurons by facilitating the clustering of L1. H2 can also be used for ELISA, WB and IF studies.

Immunofluorescence staining of fixed Neuro-2a cells with anti-NCAM-L1 antibody H2 scFv (Ab00744) Immunofluorescence analysis of paraformaldehyde fixed Neuro-2a cells on Shi-fix™ coverslips stained with the chimeric rabbit IgG version of H2 scFv (Ab00744-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00744-23.0, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (Ab00178-1.1) followed by staining with Alexa Fluor® 488 secondary antibody.
Flow cytometry using the Anti-NCAM-L1 antibody H2 scFv (Ab00744). Paraformaldehyde fixed Neuro-2a cells were stained with anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of H2 scFv (Ab00744-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
