Recombinant monoclonal antibody to CD90. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma T11D7e.

Immunogen: This antibody was raised by immunising CBA mice with xenogeneic rat thymocytes, which express the Thy-1.1 antigen.
Specificity: This antibody binds murine and rat CD90. While CD90 is a monomorphic determinant in rats, CD90 is polymorphic in mice. As a result, this antibody reacts with Thy1.1-expressing mice, including AKR and FVB mice, but not Thy1.2-expressing mice, such as CBA and BALB/c.
Application Notes: This antibody has been used in indirect immunofluorescence analysis of Thy-1 antigen expression in both methanol-fixed and living cell cultures (Dulbecco et al, 1979; Hasthorpe, 1980). This antibody has also been used, in conjunction with rabbit complement, to deplete Thy1-expressing fibroblasts from serum samples through complement-mediated lysis (Mahanthappa et al, 1996).

Flow-cytometry using the anti-CD90 antibody T11D7e (Ab00824) Rat splenocytes were fixed using 2% PFA, permeabilised using 0.5% Triton and stained with unimmunized rabbit IgG antibody (MOPC-21; isotype control, black line) or the rabbit IgG-chimeric version of T11D7e (Ab00824-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
Immunofluorescence staining of fixed rat splenocytes with anti-CD90 antibody T11D7e (Ab00824) Immunofluorescence analysis of paraformaldehyde fixed rat splenocytes permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of T11D7e (Ab00824-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00824-23.0, DAPI, merged channels and an isotype control.
