Recombinant monoclonal antibody to TCR beta. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the B-cell clone IARC307.

Immunogen: Derived from normal B cells in peripheral blood of a Burkitt’s lymphoma patient. Screened more than 60 culture supernatants from lymphoblastoid cell lines of normal origin, Epstein-Barr virus (EBV)-carrying Burkitt lymphomas, EBV-negative Burkitt lymphomas and EBV-converted sublines of original EBV-negative Burkitt lymphoma human B-cell lines for their capacity to bind the synthetic peptide autoantigens. One lymphoblastoid B-cell line was identified, IARC307, that produces IgM kappa antibodies which specifically react with the CDR1 peptide corresponding to human Vbeta8.1 gene sequence.
Specificity: This antibody is specific for Vbeta8.1 CDR1 sequence CKPISGHNSLFQWYRQT.
Application Notes: IARC307 has been used in ELISAs to bind to synthetic peptides of CRD1 region from several TCR Vbeta families of man and mouse (Marchalonis 2000).

Immunofluorescence staining of Jurkat cells with anti-TCR beta (Ab00945) IARC307 Immunofluorescence analysis of paraformaldehyde fixed Jurkat cells on Shi-fix™ coverslips stained with the chimeric rabbit IgG version of IARC307 (Ab00945-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00945-23.0, DAPI, merged channels and an isotype control. Flow cytometry using the Anti-TCR beta antibody IARC307 (Ab00945). Paraformaldehyde fixed Jurkat cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of IARC307 (Ab00945-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
