Recombinant monoclonal antibody to Fusarium. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from phage display FvCA4.
Immunogen: White leghorn chickerns were immunised by intramuscular injection of a preparation of cell wall-bound proteins isolated from the wh1-2 fungus strain of F. verticilliodes. Splenocytes were obtained from immunised chickens and used for RNA extraction and cDNA synthesis.
Specificity: FvCA4 was shown to exhibit a high affinity for F. verticilliodes cell wall-bound proteins using ELISA with both soluble antibody fragment or phage-expressed antibody (Hu et al, 2012). Again shown by ELISA, FvCA4 cross-reacted with several other Fusarium species, but not with non-Fusarium fungi.
Application Notes: FvCA4 was used in ELISAs, both as soluble scFv fragments and as phage-expressed antibodies, for the detection of purified F. verticillioides cell wall-bound proteins and Fusarium species from contaminated maize grains. In addition, cell wall-bound proteins were also detected by immunoblotting from several fungal species, not limited to the Fusarium genus. Immunofluorescent labelling showed that FvCA4 localised to the cell surface of F. verticillioides conidiospores and hyphae.
