Recombinant monoclonal antibody to GRB2. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the B-cell cDNA library SAIC-19D-1F4.
UniProt Accession Number of Target Protein: P62993
Alternative Name(s) of Target: Growth factor receptor-bound protein 2, ASH
Immunogen: Peptide "FGNDVQHFK", derived from the Growth factor receptor-bound protein 2, conjugated to KLH.
Specificity: Recognises human GRB2.
Application Notes: Original data characterizing this antibody may be found here .
Antibody first published in:
Schoenherr et al. Anti-peptide monoclonal antibodies generated for immuno-multiple reaction monitoring-mass spectrometry assays have a high probability of supporting Western blot and ELISA. Mol Cell Proteomics PMID:25512614
Note on publication:
Describes generation a panel of recombinant antibodies using synthetic peptides of human proteins as well as validation using ELISA and multiple reaction monitoring-mass spectrometry.
Western Blot using anti-GRB2 antibody SAIC-19D-1F4 (Ab00310) Human spleen lysate samples (35µg protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of SAIC-19D-1F4 (Ab00310-23.0) at 0.001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for GRB2 is 25.2kDa. Ab00310-23.0 successfully detected GRB2 in human spleen lysate.
Immunofluorescence staining of fixed HeLa cells with anti-GRB2 antibody SAIC-19D-1F4 (Ab00310) Immunofluorescence analysis of paraformaldehyde fixed HeLa cells on Shi-fix™ coverslips, permeabilized with 0.15% Triton and stained with the chimeric rabbit IgG version of SAIC-19D-1F4 (Ab00310-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (1 µg/ml), showing nuclear staining. The nuclear DNA stain is DAPI (blue). Panels show from left-right, top-bottom Ab00310-23.0, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody (Ab102-23.0) followed by Alexa Fluor® 488 secondary antibody.
