Immunogen: The original antibody was raised by immunizing a mouse with CD137.
Specificity: This antibody is specific for human CD137.
Application Notes: This antibody stimulates T-cell activity (Son et al, 2004; pmid:15087232). ELISA was performed using this antibody on E.coli expressed human 4-1BB (Son et al, 2004; pmid:15087232). Mixed lymphocyte reaction was investigated with fresh peripheral blood mononuclear cells using this antibody (Son et al, 2004; pmid:15087232). This antibody was used for flow cytometry on a subline of human Jurkat 8-1 cells (Son et al, 2004; pmid:15087232). This antibody promoted long-term proliferation of PBMC (Son et al, 2004; pmid:15087232).

The humanization of agonistic anti-human 4-1BB mAb, BBK-4, using a phage display library.
Western Blot using anti-4-1BB antibody BBK4 (Ab02446). Human peripheral blood monocytes(A) (0.3µg/ml) cells lysate and human tonsil(B) (1µg/ml) tissue lysate (35µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of BBK4 (Ab02446-23.0) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunofluorescence staining of human peripheral blood leukocytes with anti-4-1BB (Ab02446) BBK4 Immunofluorescence analysis of paraformaldehyde fixed human peripheral blood leukocytes stained with the chimeric rabbit IgG version of BBK4 (Ab02446-23.0) (1:1000 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1500 dilution), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab02446-23.0, DAPI, merged channels and an isotype control. The isotype control was anti-fluorescein antibody (Ab00102-23.0) followed by staining with Alexa Fluor® 488 secondary antibody.
