Recombinant monoclonal antibody to CD4. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma 5A8.

Immunogen: The original antibody was generated by immunizing Balb/c mice with CHO cells expresssing full-length human CD4 on the cell surface.
Specificity: This antibody is specific for nonreduced CD4. Human V1 residues (through amino acid 83) are not required for efficient 5A8 binding. human residues of the rod-like B strand which connects V1 and V2 (AA83 -AA105) significantly influence 5A8 binding. human CD4 V2 residues AA105 -AA131 (strands B and C of V2) are absolutely required for 5A8 binding.
Application Notes: This antibody was used for a radioimmunoassay on human rsCD4 (US5871732). It was tested whether this antibody can block HIV-induced syncytia formation (US5871732) (Burkly et al, 1992; pmid:1380539). Western blot was used on different epitopes of CD4 using this antibody (US5871732). Immunohistochemistry was performed on human tissue with a humanized version of this antibody (Boon et al, 2002; pmid:11893418). ELISA with this antibody was used to detect bound sCD4 (Moore et al, 1992; pmid:1378510).

A novel anti-CD4 mAb that does not block CD4/gp120 binding, but that does efficiently block both viral infection and cell-cell syncytia formation, and define its contact site as residues in CD4 D2 using both mouse/human CD4 chimeras and CD4 substitution mutants is reported.
