Recombinant monoclonal antibody to CD4. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma YNB46.1.8 (Campath-9H).
UniProt Accession Number of Target Protein: P01730
Alternative Name(s) of Target: Cluster of differentiation 4; T-cell surface glycoprotein CD4; T-cell surface antigen T4/Leu-3
Immunogen: Rat T-cell line (NB2) transfected with the human CD4.
Specificity: Recognizes the 55kDa human CD4 cell surface antigen, expressed by helper T lymphocytes and by monocytes.
Antibody first published in:
Mathieson PW1, Cobbold SP, Hale G, Clark MR, Oliveira DB, Lockwood CM, Waldmann H. Monoclonal-antibody therapy in systemic vasculitis. N Engl J Med. 1990 Jul 26;323(4):250-4. PMID:2366834
Note on publication: Describes the production of the clone YNB46.1.8 and explores its therapeutic potential as a combinatorial agent with Campath 1-H for the treatment of intractable systemic vasculitis
Western Blot using anti-CD4 antibody Ab00167 Mouse thymus (lane 1) and mouse spleen (lane 2) were resolved on a 10% SDS PAGE gel and blots probed with Ab00167-10.0 at 1.5 µg/ml before being detected by a secondary antibody. The expected band size (51 kDa) is seen along with bands corresponding to dimers.
Flow-cytometry using anti-CD4 antibody YNB46.1.8 (Campath-9H, Ab00167) Human lymphocytes were stained with an isotype control (Ab00102-23.0, panel A) or the rabbit-chimeric version of YNB46.1.8/Campath-9H (Ab00167-23.0, panel B) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a AF488 conjugated donkey anti-rabbit antibody (ab150073) and cells analysed on a FACSCanto flow-cytometer.
Immunohistochemical staining of rabbit spleen using anti-CD4 antibody (Ab00167) YNB46.1.8 (Campath-9H) Anti-CD4 staining of formaldehyde fixed paraffin embedded rabbit spleen tissue, at 40x magnification. The rat IgG1-chimeric version of YNB46.1.8 (Ab00167-6.1) was used to stain samples at a concentration of 5 µg/ml.
