Immunogen: OX-45 was prepared by immunizing BALB/c mice with rat T blasts prepared by stimulating purified T helper (TH) cells with allogeneic irradiated rat spleen cells.
Specificity: OX-45 binds specifically to CD48 on rat thymocytes, T and B cells and endothelium but not LFA.3. Directly competes with CD46 for binding the target epitope. CD48 is a CD2 ligand and is expressed on the surface of all hematopoietic cells and endothelium. May be involved in the facilitation of interactions between activated lymphocytes. Likely to be involved in T cell activation.
Application Notes: Describes the use of OX-45 in blocking the primary mixed lymphocyte response (MLR)and the secondary response of T lymphocytes to the soluble antigen DNP-BGG. It also showed that OX-45 did not have any effect on the cytotoxic activity of specific (CTL) or non-specific (NK) killer cells or on proliferative responses stimulated by lectins or oxidative mitogenesis.

Flow-cytometry using anti-CD48 antibody OX-45 (Ab00571) Staining of rat (Rattus norvegicus) lymphocytes with either an isotype control (A) or the rabbit-chimeric version of OX-45 (B, Ab00571-23.0) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using an Alexa Fluor ® 488 conjugated donkey anti-rabbit antibody (ab150073) and cells analysed on a FACSCanto flow-cytometer.
Western Blot using anti-CD48 antibody OX-45 (Ab00571) Rat spleen (A), thymus (B) and lymph node (C) tissue lysate samples (35µg protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of OX-45 (Ab00571-23.0) at 0.001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for CD48 is ~37-56kDa and it is heavily glycosylated as well as lipidated (UniProt ID: P10252). Ab00571-23.0 successfully detected CD48 in rat spleen, thymus and lymph node tissue lysates.
