Immunogen: The original antibody was generated by immunizing BALB/c mice with human cytokeratin 18 extracellular domain polypeptide 'LEDGEDFNLGDALD'. Later on the humanized version of this antibody was generated by grafting the CDRs of the original mouse antibody E156 onto human framework regions.
Specificity: This antibody is specific for extracellular domain of the human cytokeratin 18 containing amino acids 'LEDGEDFNLGDALD'. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. Cytokeratin 18 is widely expressed in human monolayer epithelial tissues, and generally appears in pairs with CK8. CK18 is a serum marker for a variety of tumor diagnosis, as well as a serum marker for liver injury. It is involved in the uptake of thrombin-antithrombin complexes by hepatic cells. When phosphorylated, it plays a role in filament reorganization. It is also involved in the delivery of mutated CFTR to the plasma membrane. Together with KRT8, is involved in interleukin-6 (IL-6)-mediated barrier protection.
Application Notes: This antibody can bind human cytokeratin 18 in an ELISA. The original IgG1 version of this antibody is reported to bind human cytokeratin 18 with a binding affinity of 32.8 pM.

Flow cytometry using anti-Cytokeratin 18 antibody HH156 (Ab03185). Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with the anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of HH156 (Ab03185-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.
