Immunogen: The original antibody was isolated from a VHH library generated by immunization of an alpaca with whole glioblastoma multiforme (GBM) cells.

Specificity: This antibody binds human dihydropyrimidinase-related protein 2 (DPYSL2). This protein plays a role in neuronal development and polarity, as well as in axon growth and guidance, neuronal growth cone collapse and cell migration. It is necessary for signaling by class 3 semaphorins and subsequent remodeling of the cytoskeleton and may play a role in endocytosis. This antibody was also reported to cross react with human methylenetetrahydrofolate dehydrogenase 1 (MTHFD1), a trifunctional enzyme that catalyzes the interconversion of three forms of one-carbon-substituted tetrahydrofolate.

Application Notes: This antibody was used for identification of DPYSL2 and MTHFD1 proteins in cytosolic and membrane protein fractions of glioblastoma (GBM) tissues using western blotting. It was reported that DPYSL2 expression was high in GBM compared to reference samples. ELISA was used for the screening of the nanobody library and identification of specific binders that showed specificity for GBM proteins (PMID: 28498803).
