Recombinant monoclonal antibody to dT5. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the phage display antibody DNA-1.

Immunogen: The antibody was isolated from a bacteriophage Fab display library derived from autoimmune (MRL/lpr) mice. The library was panned against single-stranded (ss) calf thymus (CT) DNA and the Fabs was selected.
Specificity: The antibody binds to dT5, primarily recognising the thymine bases of DNA. The antibody binds also to oligo(dT)15 nucleotides or greater as well as oligo(dT) of length three to five nucleotides. The antibody also recognises the abbreviate hairpin sequence LIG5-14.
Application Notes: The crystal structure of the Fab fragment in complex with dT5 was determined (Tanner et al., 2001; PMID: 11733999). Fluorescence titration of the antibody with dT5 was measured, with a Kd of approximately 27 µM (Komissarov et al., 1996). Isothermal titration calorimetry experiments were used to measure the apparent affinity of the antibody for dT5. The apparent association constant was KA = 6.0*10^4 M-1. Further, the binding of dT5 to the antibody was exothermic, with an apparent ΔHcal under our solution conditions of -16.8 kcal/mol (Schuermann et al., 2004; PMID: 15340914).
