Immunogen: The original version of this antibody was raised by Epstein-Bar virus (EBV) transformation of peripheral blood mononuclear cells obtained from HIV-infected subjects followed by fusion with the SHM-D33 heteromyeloma (Buchbinder et al., 1992; pmid: 1466965).
Specificity: 447-D recognizes a neutralizing epitope of the hypervariable V3 loop of HIV-1 gp120. In the loop V3, it recognizes the highly conserved GPGR sequence present in the β-turn of V3 of many HIV strains. It is suggested that the antibody 447-52D neutralizes HIV-1 MN by capturing the V3 loop, blocking its interaction with the coreceptor and altering the structure of the envelope spike (Dutta et al., 2014; pmid: 25122783).
Application Notes: This antibody was demonstrated to be neutralizing against HIV (Buchbinder et al., 1992; pmid: 1466965). It can be used synergistically with other neutralizing HIV antibodies (such as 694-D). Although specific to the V3 loop of gp120 protein, this clone was shown to bind to various HIV isolates, such as MN, RF or HTLV (Buchbinder et al., 1992; pmid: 1466965). This clone also neutralizes both X4 and R5 primary isolates, making it one of the most effective anti-V3 antibodies characterized to date. Consequently, this antibody is recommended for research on the develpment of successful treatment and prevention of HIV infection, as effective neutralizing antibodies are capable of depleting the viral load in patients exposed to the virus.
