Recombinant monoclonal antibody to IgG. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma NH3/130.5.2.
Alternative Name(s) of Target: immunoglobulin G; human immunoglobulin; NH3/130
Immunogen: This antibody was raised by immunising Balb/c mice with whole IgG fraction and subsequent hybridoma cells formation.
Specificity: This antibody recognises human IgG. The antibody cross-reacts with rhesus macaque IgG1, cynomolgus monkey IgG1 and cynomolgus monkey IgG4.
Application Notes: This antibody was shown to have a high avidity to human immunoglobulin G having a high titre, equal to a corresponding rabbit polyclonal antibody, when tested against anti-D sensitised cells (Downie et al., 1983). NH3/130.5.2 detects human IgG samples transferred from the SDS polyacrylamide gel via Western blot under non-reducing conditions (Thorpe et al., 1984). Thorne et al. (1984) mentions that this antibody was also successful in immunoprecipitation in agar or solution (M. Spitz, unpublished results). Further, the antibody was capable of detecting IgGs via Western blot from native gels run under acidic conditions (Thorne et al., 1984). Finally, the antibody cross-reacted with rhesus macaque IgG1, cynomolgus monkey IgG1 and cynomolgus monkey IgG4 as shown by ELISA analysis.

This article describes the generation and characterisation of the NH3/130.5.2 antibody.
ELISA using NH3/130.5.2 (Ab00854) and Rhesus macaque IgG1, Cynomolgus monkey IgG1, Cynomolgus monkey IgG4 and Human IgG1. Binding curves of the mouse IgG1 chimeric version of the Anti-IgG antibody NH3/130.5.2 (Ab00854-1.1; orange line) and isotype control (Ab00102-1.1, anti-fluorescein; dotted orange line) to an ELISA plate coated with Rhesus macaque IgG1 (Ab00102-26.0, Absolute Antibody) at a concentration of 2.5 ug/ml, of the mouse IgG1 chimeric version of the Anti-IgG antibody NH3/130.5.2 (Ab00854-1.1; blue line) and isotype control (Ab00102-1.1, anti-fluorescein; dotted blue line) to an ELISA plate coated with Cynomolgus monkey IgG1 (Ab00102-35.0, Absolute Antibody) at a concentration of 2.5 ug/ml, of the mouse IgG1 chimeric version of the Anti-IgG antibody NH3/130.5.2 (Ab00854-1.1; purple line) and isotype control (Ab00102-1.1, anti-fluorescein; dotted purple line) to an ELISA plate coated with Cynomolgus monkey IgG4 (Ab00102-36.0, Absolute Antibody) at a concentration of 2.5 ug/ml and of the mouse IgG1 chimeric version of the Anti-IgG antibody NH3/130.5.2 (Ab00854-1.1; green line) and isotype control (Ab00102-1.1, anti-fluorescein; dotted green line) to an ELISA plate coated with Human IgG1 (Ab00102-10.0, Absolute Antibody) at a concentration of 2.5 ug/ml. A 3-fold serial dilution from 3000 to 0.0169 ng/ml was performed using Ab00854-1.1 and an anti-mouse IgG was used as secondary antibody for detection.
