Recombinant monoclonal antibody to IL-18. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the B-cell cDNA library SAIC-24B-4.
UniProt Accession Number of Target Protein: Q14116
Alternative Name(s) of Target: Interleukin-18, IGIF, IL1F4
Immunogen: The KLH conjugated peptide "ISTLSCENK" derived from IL-18, an augmentor of natural killer cell activity in the spleen and stimulator of interferon gamma production in Th1 cells.
Specificity: Recognises human IL18.
Application Notes: Original data characterizing this antibody may be found here .
Antibody first published in:
Schoenherr et al. Anti-peptide monoclonal antibodies generated for immuno-multiple reaction monitoring-mass spectrometry assays have a high probability of supporting Western blot and ELISA. Mol Cell Proteomics PMID:25512614
Note on publication: Describes generation a panel of recombinant antibodies using synthetic peptides of human proteins as well as validation using ELISA and multiple reaction monitoring-mass spectrometry.
Western Blot using anti-IL-18 antibody SAIC-24B-4 (Ab00316) Untreated human peripheral blood monocyte (PBM; A) and PMA/lonomycin-treated PBM lysates (35µg protein in RIPA buffer) were resolved on a 15% SDS PAGE gel and blots probed with the rabbit IgG-chimeric version of SAIC-24B-4 (Ab00316-23.0) at 0.3 µg/ml before detection using an anti-rabbit secondary antibody. Protein was detected by chemiluminescence. The expected running size for IL-18 is 22.3kDa. Ab00316-23.0 successfully detected IL-18 in untreated human peripheral blood monocyte samples.
