Recombinant monoclonal antibody to Lewis a,b. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma HEA164.
Alternative Name(s) of Target: Blood Group Lewis a/b; lewis a; lewis b
Immunogen: This antibody was raised by immunising mice with human carcinoma cells.
Specificity: This antibody is specific for Lewis a,b carbohydrate antigens.
Application Notes: This antibody has applications in the detection of normal and neoplastic epithelial cells. This antibody has been shown to react with a glycolipid fraction isolated from colon carcinoma tissue, with no effect of pronase E addition on its reactivity (Moldenhauer et al, 1987). This antibody, when immobilised on the wall of a shear chamber, has been shown to support slow leukocyte rolling (Chen et al, 1997).

Western Blot using anti-Lewis a,b antibody HEA164 (Ab01422) Human colorectal carcinoma tissue lysate (35µg protein in RIPA buffer) was resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of HEA164 (Ab01422-23.0) at 0.001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunofluorescence staining of fixed Caco-2 cells with anti-Lewis a,b antibody HEA164 (Ab01422) Immunofluorescence analysis of paraformaldehyde fixed Caco-2 cells on Shi-fix™ coverslips stained with the chimeric rabbit IgG version of HEA164 (Ab01422-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The isotype control was an unknown specificity antibody (Ab00178-23.0) followed by staining with Alexa Fluor® 488 secondary antibody.
