Immunogen: This antibody was raised by immunising mice with red cell membranes.
Specificity: This antibody recognises human Spectrin which is a cytoskeletal protein creating scaffolding of the plasma membrane.
Application Notes: IHC: RG/26 antibody was used to stain membranes of iRBCs infected with a parasite in a study which investigated influence of vascular endothelial growth factor on red cells infected with Plasmodium falciparum (Sachanonta et al., 2008). WB: This antibody was also used in Western blot analysis to assess free and degraded spectrin as an element of a study which aimed at investigating the infuence of constant storage temperatures and multiplewarming cycles on stored red blood cells (Wagner et al., 2014).

Flow cytometry using the Anti-Spectrin antibody RG/26 (Ab01319). Paraformaldehyde fixed K562 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of RG/26 (Ab01319-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
Western Blot using anti-Spectrin antibody RG/26 (Ab01319). Human red blood cell (RBC) lysates (35µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of RG/26 (Ab01319-23.0) at 0.001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunofluorescence staining of K562 cells with anti-Spectrin (Ab01319) RG/26 Immunofluorescence analysis of paraformaldehyde fixed K562 cells on Shi-fix™ cover-slips stained with the chimeric rabbit IgG version of RG/26 (Ab01319-23.0) (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing membrane and cytoplasmic staining.
