Immunogen: This antibody was raised by immunising mice with CD16 antigen.
Specificity: This antibody recognises an epitope in the first membrane-distal domain of both CD16a and CD16b. CD16 is a low affinity receptor for IgG, and plays an important role in mediating ADCC by NK cells.
Application Notes: DJ130c may be used in identification and characterisation of various cancers. For instance, one group utilised this antibody in the analysis of expression profiles of gastric cancer xenografts exposed to cisplatin and considerable alteration in the expression level of CD16 was observed (Cai et al., 2017). Another group used this antibody to identify and compare different subpopulations of macrophages and their potential relation to acute heart transplant rejection, both via immunofluorescence and immunohistochemistry in tissue samples (van den Bosch et al., 2017).

Western Blot using anti-CD16 antibody DJ130c (Ab01342). human peripheral blood leukocytes(A) (0.03 µg/ml), human spleen(B) (0.01 µg/ml), human tonsil(C) (0.3 µg/ml) and human placenta(D) (0.03 µg/ml) tissue lysate (35µg protein in RIPA buffer) was resolved on an SDS PAGE gel and blots probed with the chimeric mouse IgG version of DJ130c (Ab01342-1.1) before detection using an anti-mouse secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunofluorescence staining of U937 cells using anti-CD16 (Ab01342) DJ130c Immunofluorescence analysis of paraformaldehyde fixed U937 cells stained with the chimeric mouse IgG version of DJ130c (Ab01342-1.1) at 10 µg/ml followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01215-1.1, DAPI, merged channels and an isotype control. The isotype control was stained with anti-unknown antibody (Ab00178-1.1) followed by Alexa Fluor® 488 secondary antibody.

Flow cytometry using the Anti-CD16 antibody DJ130c (Ab01342). Paraformaldehyde fixed human peripheral blood monocytes cells were stained with anti-unknown specificity antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG1 version of DJ130c (Ab01342-1.1 , blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-mouse IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
