Recombinant monoclonal antibody to CD21L. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma R4/23.

Immunogen: This antibody was raised by immunising BALB/c mice with C3 receptor-coated erythrocytes.
Specificity: This antibody recognises CD21L which is a long isoform of CD21 (CR2) (with an additional exon 10a). It is strongly expressed on dendritic reticulum cells and serves as a receptor for the complement elements (and is also bound by EBV).
Application Notes: CD21L is strongly expressed in reticulum dendritic cells and R4/23 might be used for the identification of these cells. For instance, Schriever et al. (1989) used this antibody (among others) in an attempt to differentiate follicular dendritic cells from lymphoid dendritic cells. This antibody can be also utilised in the characterisation of certain cancers; for example, one group used it in an immunohistochemical analysis of frozen samples of tumour cells from diffuse large B‐cell lymphoma (Ogawa et al., 2004).

Immunofluorescence staining of fixed Daudi cells with anti-CD21L antibody R4/23 (Ab01339) Immunofluorescence analysis of paraformaldehyde fixed Daudi cells on Shi-fix™ coverslips stained with the chimeric rabbit IgG version of R4/23 (Ab01339-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01339-23.0, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (Ab00178-1.1) followed by staining with Alexa Fluor® 488 secondary antibody.
Flow cytometry using the Anti-CD21L antibody R4/23 (Ab01339). Paraformaldehyde fixed Daudi cells were stained with anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of R4/23 (Ab01339-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
