Immunogen: This antibody was raised in mice against RFLPADQMNVADPQPLASCS peptide sequence of human CD4.
Specificity: This antibody recognises the amino-acid sequence corresponding to the 3'-UGA codon region of human CD4 (downstream of the natural UGA (STOP) codon transcribed, if termination at the UGA codon is suppressed). CD4 is a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigenes and is also a receptor for the human immunodeficiency virus.
Application Notes: 8A5 it reacts with the region of the human CD4 gene downstream of the natural UGA (STOP) codon when it is transcribed (UGA as a selenocysteine), and thus this antibody is recommended for studies of the role of selenium metabolism and selenoproteins in diseases, such as infectious diseases and cancer (Ware, 2003).

Western Blot using anti-CD4 antibody 8A5 (Ab01464). U937(A) (2µg/ml) cells lysate and human spleen(B) (2µg/ml) tissue lysate (35µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of 8A5 (Ab01464-23.0) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunofluorescence staining of human peripheral blood leukocytes with anti-CD4 (Ab01464) 8A5 Immunofluorescence analysis of paraformaldehyde fixed human peripheral blood leukocytes stained with the chimeric rabbit IgG version of 8A5 (Ab01464-23.0) (1:1000 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1500 dilution), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01464-23.0, DAPI, merged channels and an isotype control. The isotype control was anti-fluorescein antibody (Ab00102-23.0) followed by staining with Alexa Fluor® 488 secondary antibody.
