Immunogen: This antbody was raised by immunising mice with phytohaemagglutinin-activated peripheral blood mononuclear cells.
Specificity: This antibody recognises human CD68 antigen. CD68 is a membrane glycoprotein highly expressed in monocytes and macrophages. It is involved in phagocytosis, lysosomal metabolism and interactions with pathogens and other cells.
Application Notes: IP: Y-2/131/47 antibody was used for immunoprecipitation from the material from spleen membranes (Micklem et al., 1989). ICC: This antibody was also used in immunocytochemical staining of acute promyelocytic leukaemia cells in order to establish a characteristic phenotype of the diseased cells and develop a potential diagnostic tool of this disorder (Erber et al., 1994).

Immunofluorescence staining of K562 cells using anti-CD68 (Ab01316) Y-2/131/47 Immunofluorescence analysis of paraformaldehyde fixed K562 cells stained with the chimeric mouse IgG version of Y-2/131/47 (Ab01316-1.1) at 10 µg/ml followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane and cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01215-1.1, DAPI, merged channels and an isotype control. The isotype control was stained with anti-unknown antibody (Ab00178-1.1) followed by Alexa Fluor® 488 secondary antibody.
Flow cytometry using the Anti-CD68 antibody Y-2/131/47 (Ab01316). Paraformaldehyde fixed U937 cells were stained with anti-unknown specificity antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG1 version of Y-2/131/47 (Ab01316-1.1, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-mouse IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
