Recombinant monoclonal antibody to CD9. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma P1/33/2.

Immunogen: This antibody was raised by immunising mice with acute lymphoblastic leukaemia cells.
Specificity: This antibody recognises CD9 which is a glycoprotein with four transmembrane domains that is expressed on the surface of various blood cells, such as B lymphocytes, platelets, monocytes and others, and also on neurons and glia on the periphery. It is implicated in different functions, for example cell-cell adhesion, signal transduction and platelet activation.
Application Notes: Antibodies against CD9 might be used in characterisation of acute leukaemias, other tumours and for functional studies on platelets. For instance, one group used P1/33/2 during immunohistochemical characterisation of frozen sections of melanoma ABCG2+IGR37 cells xenografts (Taghizadeh et al., 2010). Another group used this antibody (along with other anti-CD9 antibodies) to analyse CD9 expression level and distribution in different breast cancer cells (MDA, MA-11, and MCF-7) and they showed that CD9 might be a useful marcer of the neoplastic cells as its expression was higher in cancer cells than in normal breast cells as well as it was additionally present in the nuclei of the malignant tissue (Rappa et al., 2014).

Immunofluorescence staining of human peripheral blood leukocytes using anti-CD9 (Ab01340) P1/33/2 Immunofluorescence analysis of paraformaldehyde fixed human peripheral blood leukocytes stained with the chimeric mouse IgG version of P1/33/2 (Ab01340-1.1) at 10 µg/ml followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01215-1.1, DAPI, merged channels and an isotype control. The isotype control was stained with anti-unknown antibody (Ab00178-1.1) followed by Alexa Fluor® 488 secondary antibody.
Flow cytometry using the Anti-CD9 antibody P1/33/2 (Ab01340). Paraformaldehyde fixed human peripheral blood monocytes cells were stained with anti-unknown specificity antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG1 version of P1/33/2 (Ab01340-1.1, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-mouse IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
