Immunogen: The original antibody was generated by immunizing female BALB/c mice with an immunogenic polypeptide containing amino acids 410-432 (TIVEEVQDGKVISSREQVHQTTR) of human cytokeratin 17 conjugated with KLH.
Specificity: This antibody is specific for amino acids 410-432 (TIVEEVQDGKVISSREQVHQTTR) of human cytokeratin 17. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. Cytokeratin 17 regulates the activity of kinases such as PKC and SRC via binding to integrin beta-1 (ITB1) and the receptor of activated protein C kinase 1 (RACK1).
Application Notes: The binding characterization of this antibody to human cytokeratin 8 was done using ELISA. The affinity constant for the original IgG3 antibody was calculated to be 9.6 × 10^(8) M^(-1). This antibody is similar to commercially available anti-Cytokeratin 17 antibody E3 in staining various human tissues. This antibody can be used in the immunohistochemical analysis of esophageal squamous cell carcinoma (CN113278070).

Flow cytometry using anti-Cytokeratin 17 antibody 4D2 (Ab03182). Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with the anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of 4D2 (Ab03182-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.
Western blot using anti-Cytokeratin 17 antibody 4D2 (Ab03182). A431 (A) (0.001µg/ml) and HeLa (B) (0.0001µg/ml) cells lysate (35µg protein in RIPA buffer) were resolved on an SDS-PAGE gel, and blots were probed with the chimeric rabbit version of 4D2 (Ab03182-23.0) at the mentioned respective concentrations before detection using an anti-rabbit secondary antibody. A primary incubation of 1 hour was used, and protein was detected by chemiluminescence.
