Immunogen: The original antibody was generated by immunizing female ICR mice with recombinant human CK20 histidine-tagged fusion protein in Freund's complete adjuvant.
Specificity: This antibody binds human cytokeratin 20. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. There are 19 keratin polypeptides are found in human epithelium. Cytokeratin 20 plays a significant role in maintaining keratin filament organization in intestinal epithelia. When phosphorylated, it plays a role in the secretion of mucin in the small intestine.
Application Notes: The binding characterization of this antibody was done using ELISA. The affinity constant of the original IgG2a antibody was determined to be 1.92 × 10^9 M^(-1). This antibody was capable of detecting recombinant CK20 protein in a western blot assay. This antibody is similar to commercially available anti-Cytokeratin 20 antibody KS20.8 in staining various human tissues. This antibody was used in the immunohistochemical staining of stomach epithelial tissues and gastric adenocarcinomas (CN113583120).

Flow cytometry using anti-Cytokeratin 20 antibody 10A2 (Ab03190). Paraformaldehyde fixed Caco-2 cells permeabilized with 0.5% Triton were stained with the anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of 10A2 (Ab03190-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.
Western blot using anti-Cytokeratin 20 antibody 10A2 (Ab03190). Human duodenum (A) (0.01µg/ml), ileum (B) (0.001µg/ml), and colon (C) (0.001µg/ml) tissue lysate (35µg protein in RIPA buffer) were resolved on an SDS-PAGE gel, and blots were probed with the chimeric rabbit version of 10A2 (Ab03190-23.0) at the mentioned respective concentrations before detection using an anti-rabbit secondary antibody. A primary incubation of 1 hour was used, and protein was detected by chemiluminescence.
Immunofluorescence staining of Caco-2 cells with anti-Cytokeratin 20 antibody 10A2 (Ab03190).
