Immunogen: CV5.1 antibody was raised in mouse using His-tagged denatured FANCM (aa 1507-1679) made in E coli as the immunogen.
Specificity: This antibody recognises human FANCM (other species were not tested but there is high sequence conservation). FANCM is a part of a larger FA (Fanconi anemia) complex. It is engaged in DNA repair, resistance to DNA cross-linking drugs and prevention of chromosomal breakage.
Application Notes: IHC: CV5.1 antibody successfully detected FANCM in human fetal and adult ovaries (Fouquet et al., 2017). WB: Fouquet et al. (2017) also performed western blot analysis detecting FANCM with this antibody. IHC: FANCM in human testicular sections was visualised by the CV5.1 antibody (Kasak et al., 2018).

This article describes the earliest found use of the CV5.1 antibody, both in immunohistochemistry and immunoblotting.
Immunofluorescence staining of HeLa cells using anti-FANCM antibody (Ab01300) CV5.1. Immunofluorescence analysis of paraformaldehyde fixed HeLa cells permeabilized with 0.15% Triton and stained with the chimeric mouse IgG version of CV5.1 (Ab01300-1.1) at 10 µg/ml followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing nuclear staining. Actin filaments were stained with phalloidin (red) and the nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab01300-1.1, DAPI, merged channels and an isotype control. The isotype control was stained with anti-unknown antibody (Ab00178-1.1) followed by Alexa Fluor® 488
Flow-cytometry using the anti-FANCM antibody CV5.1 (Ab01300). HeLa cells were fixed using paraformaldehyde, permeabilised using 0.5% Triton and stained with anti-unknown IgG antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG-chimeric version of CV5.1 (Ab01300-1.1, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-mouse IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
