Recombinant monoclonal antibody to Histone H4. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma 1A9D7.

Immunogen: The original antibody was generated by immunizing mice with Histone H4 protein with site-specific acetylations at K5 and K8.
Specificity: The antibody specifically recognizes simultaneous acetylations at K5 and K8 of histone H4. Lysine acetylation of core histones is one of the major posttranslational modifications involved in the control of eukaryotic gene expression.
Application Notes: The specificity of the original format of the antibody for histone H4 proteins containing di-acetylation of K5acK8ac was confirmed by ELISA analysis. The antibody detected the histone H4 proteins containing at least the di-acetylation of K5acK8ac western blot analysis. The crystal structure of the complex of di-acetylated histone H4 and 1A9D7 Fab fragment was determined (Handoko et al., 2018; PMID: 30080437).

Immunofluorescence staining of HeLa cells with anti-Histone H4 antibody 1A9D7 (Ab03227). Immunofluorescence analysis of paraformaldehyde fixed HeLa cells, permeabilized with 0.15% Triton, on Shi-fix™ coverslips stained with the chimeric rabbit IgG version of 1A9D7 (Ab03227-23.0) (1:100 dilution) for 1 hour followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing nuclear staining. Actin filaments were stained with phalloidin (red), and the nuclear stain is DAPI (blue). Panels show, from left to right, top to bottom, Ab03227-23.0, DAPI, merged channels, and an isotype control. The isotype control was an unknown specificity antibody (Ab00178-23.0) followed by staining with Alexa Fluor® 488 secondary antibody.
