Recombinant monoclonal antibody to Ki-67. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma 585.

Immunogen: This antibody was raised by immunizing mice with recombinant Ki-67 protein.
Specificity: This antibody recognizes Ki-67, a prominent proliferation marker present during cell division.
Application Notes: 585 can be used for the identification of Ki-67. Ki-67 protein is present during all active phases of the cell cycle (G(1), S, G(2), and mitosis), but is absent from resting cells (G(0)), what makes it an excellent marker for determining the so-called growth fraction of a given cell population (Scholzen and Gerdes, 2000; pmid: 10653597). Detection of Ki-67 is also widely used as an important marker of certain conditions, such as breast cancer (Inwald et al., 2013; pmid: 23674192).

Flow cytometry using the Anti-Ki-67 antibody 585 (Ab01429). Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG1 version of 585 (Ab01429-1.1, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
Immunofluorescence staining of HeLa cells with Anti-Ki-67 (Ab01429) 585 Immunofluorescence analysis of paraformaldehyde fixed HeLa cells permeabilized with 0.15% Triton stained with the chimeric mouse IgG1 version of 585 (Ab01429-1.1) (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing nucleoli and nucleoplasm staining.
