Immunogen: This antibody was raised against human RAP1.
Specificity: This antibody recognises human and mouse RAP1, which is a part of the shelterin complex involved in the regulation of telomere length and protection. Additionally, RAP1 acts as transcription factor at extratelomeric sites regulating gene expression.
Application Notes: WB and IHC: RAP1 was visualised multiple times in different Western blot analyses using RAP1 4C8/1 antibody; furthermore, this antibody was used to identify RAP1 immunohistochemically in invasive ductal carcinoma samples in experiments on the RAP1's role in regulating NF-kappaB (Teo et al., 2010). WB: Tascher et al. (2017) compared RAP1 protein amounts in mice skeletal muscle samples on Western blots using this antibody to investigate changes in expression of telomeric proteins during space flights. IF/ICC: RAP1 was successfully visualised in hMSCs using RAP1 4C8/1 antibody (Zhang et al., 2019).

This article describes the earliest published use of anti-RAP1 4C8/1 antibody in Western blot analyses in the experiments on the mechanisms preventing end-joining in telomeric regions.
Immunofluorescence staining of HeLa cells with anti-RAP1 antibody 4C8/1 (Ab01303). Immunofluorescence analysis of paraformaldehyde-fixed HeLa cells, permeabilized with 0.15% Triton, on Shi-fix™ coverslips stained with the Mouse IgG2b version of 4C8/1 (Ab01303-3.0) (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing nuclear staining. Actin filaments were stained with phalloidin (red), and the nuclear stain is DAPI (blue). Panels show, from left-right, top-bottom, Ab01303-3.0, DAPI, merged channels, and an isotype control. The isotype control was an unknown specificity antibody (Ab00178-3.0) followed by staining with Alexa Fluor® 488 secondary antibody.
Western blot using anti-RAP1 antibody 4C8/1 (Ab01303). NIH3T3 (A) (0.01µg/ml), HeLa (B) (0.01µg/ml), K562 (C) (0.01µg/ml), Jurkat (D) (0.01µg/ml), HEK293 (E) (0.001µg/ml), HepG2 (F) (0.003µg/ml), and A431 (G) (0.003µg/ml) cells nuclear lysates (35µg protein in RIPA buffer) were resolved via SDS-PAGE, and the subsequent blots were probed with the mouse version of 4C8/1 (Ab01303-3.0) at the mentioned respective concentrations before detection using an anti-mouse secondary antibody. A primary incubation of 1 hour was used, and proteins were detected by chemiluminescence.
