Recombinant monoclonal antibody to S100A9. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma IID4B10.

Immunogen: The original antibody was raised by immunizing Balb/c mice with human blood cells.
Specificity: This antibody is specific for the S100A9 protein and does not cross react with S100A8, S100A12 or S100A13.
Application Notes: This antibody was used for ELISA on recombinant S100A9 protein (CN104894074). Western blot was performed on S100A9, S100A8, S100A12 and S100A13 using this antibody (CN104894074). Immunohistochemistry was performed on human breast cancer, colon cancer, and hepatocellular carcinoma tissues using this antibody (CN104894074).

Western Blot using Anti-S100A9 antibody IID4B10 (Ab02468). Human PBM(A) (0.01µg/ml), human spleen(B) (0.1ug/ml), human tonsil(C) (0.001ug/ml) and human bone marrow(D) (0.001ug/ml) tissue lysates (35µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric mouse version of IID4B10 (Ab02468-1.1) before detection using an anti-mouse secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunofluorescence staining of U937 cells with Anti-S100A9 (Ab02468) IID4B10 Immunofluorescence analysis of paraformaldehyde fixed U937 cells immobilized on Shi-fix™ plus cover-slips, permeabilized with 0.15% Triton stained with the chimeric mouse IgG1 version of IID4B10 (Ab02468-1.1) (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing nuclear and cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab02468-1.1, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (Ab00178-1.1) followed by staining with Alexa Fluor® 488 secondary antibody.
Flow cytometry using the Anti-S100A9 antibody IID4B10 (Ab02468). Paraformaldehyde fixed U937 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (Ab00178-1.1; isotype control, black line) or the mouse IgG1 version of IID4B10 (Ab02468-1.1, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
