Immunogen: The original antibody was generated by immunizing BALB/c mice or SJL mice with human and mouse TIM-3-Fc protein.
Specificity: This antibody binds human TIM-3 and capable of inhibiting the interaction of TIM-3 and LILRB2. It is reported that this antibody strongly binds CC′ loop and mildly to the DE loop of TIM-3. Hepatitis A virus cellular receptor 2 (HAVCR2), also known as T-cell immunoglobulin and mucin-domain containing-3 (TIM-3), is a protein expressed on the cell surface of IFNγ producing CD4+ Th1 and CD8+ Tc1 cells. This protein is also expressed on the surface of Th17 cells, regulatory T-cells and innate immune cells like dendritic cells, NK cells and monocytes. It is a cell surface receptor implicated in modulating innate and adaptive immune responses (US20160200815).
Application Notes: This antibody can inhibit the interaction between TIM-3 and LILRB2 and can be used in the treatment of TIM-3 related disorders. The binding characterization of this antibody to human and mouse TIM-3 was done using ELISA and flow cytometry. Blocking of TIM-3:LILRB2 interactions by this antibody was evaluated in activated-macrophage and activated-DC assays. This antibody was able to initiate TNFα secretion in LPS activated macrophages. The stimulation of pro-inflammatory cytokines GM-CSF, IL-1α, IL-1β, IL-6 and TNFα by this antibody was greater for macrophages from the LILRB2+ donor compared to the low LILRB2 donor
