Recombinant monoclonal antibody to metarhodopsin-I. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma K42-41L.
UniProt Accession Number of Target Protein: P08100
Alternative Name(s) of Target: opsin-2; meta I rhodopsin; rhodopsin; opsin 2; metarhodopsin I
Immunogen: This antibody was raised by immunising mice with rhodopsin.
Specificity: This antibody recognises secifically C3 loop of metarhodopsin-I (residues 230-252) (and also dark-adapted form of rhodopsin) but it does not bind to the ligh-activated form - metarhodopsin-II. Rhodopsin is the best studied GPCR and is present in rod cells where it is activated by light being responsible for the signal transduction in the visual system.
Application Notes: Antibody imprint: This antibody was used to investigate the structure of rhodopsin's meta-states; the researchers employed the antibody imprint technique to analyse conformational changes which this molecule undergoes upon activation by light (Bailey et al., 2003). Another group used K42-41L antibody to demonstrate that it is capable of discriminating between metarhodopsins I and II, and that it stabilises metarhodopsin I while inhibiting metarhodopsin II; these data suggest that this antibody might be useful in the structural studies of opsins (Piscitelli et al., 2006). EM ICC: K42-41L antibody was utilised to stain different structures of Xenopus laevis retinas in EM immunocytochemistry LR white samples in a study investigating opsin localisation (Besharse and Wetzel, 1995).
