Immunogen: This antibody was raised by immunizing Balb/c mice with a 90 kDa MBP/alpha catenin fusion protein purified on an amylose column.

Specificity: This antibody recognizes alpha catenin and binds to the domain between amino acids 135-226. α-catenin associates with the cytoplasmic domain of a variety of plasma membrane spanning cadherins that mediate cell-cell adhesion. The association of catenins to cadherins produces a complex that is linked to the actin filament network, which seems to be of primary importance for their cell-adhesion properties.

Application Notes: This antibody is recommended for the detection and analysis of alpha-catenin via various methods. For instance, it was used to identify alpha-catenin in MDA468 cells via immunofluorescence (Bajpai et al., 2009). It was also repeatedly demonstrated that 1G5 specifically detects alpha-catenin on Western blots (Johnson et al., 1993; Stadler et al., 2018).

Western Blot using anti- alpha catenin antibody 1G5 (Ab01654) A431(A) (0.003µg/ml), HeLa(B) (0.03ug/ml) and MCF7(C) (0.01ug/ml) cell lysate. (35µg protein in RIPA buffer) was resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of 1G5 (Ab01654-23.0) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
