Recombinant monoclonal antibody to Arginase 1. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma mAb5.

Immunogen: The antibody was generated by immunizing mice with recombinant arginase 1.
Specificity: The antibody binds arginase 1. The binding of the antibody does not sterically block the Arginase 1 active site, but it interacts with an amino acid (Arg21) important to substrate binding by arginase 1, thereby inhibiting enzyme activity. Arginase is a manganese-containing enzyme that catalyzes the hydrolysis of L-arginine to L-ornithine and urea, and modulates T-cell-mediated immune response.
Application Notes: The structure and epitope mapping of the complex arginase:antibody was determined by cryo-electron microscopy. The antibody was assessed for its potency and extent of inhibition of mouse arginase using an assay that detects the formation of thioornithine from thioarginine. In this assay, as well as an LCMS assay, it was determined that the antibody could fully inhibit the enzymatic activity of arginase. To assess the kinetic mechanism of inhibition of the antibody, increasing fixed concentrations of antibody were tested across a range of thioarginine concentrations. The Ki of the antibody against mouse Arg1 was 25 ± 1.7 nM (Palte et al., 2021; PMID: 34326456).
