Recombinant monoclonal antibody to BDE-47. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the phage display antibody 7.

Immunogen: The original antibody was generated by immunizing an alpaca with a surrogate of BDE-47 covalently attached to a carrier protein. The resulting mRNA coding for the variable domain of heavy-chain antibodies were isolated, transcribed to cDNA, and cloned into a phagemid vector for phage display library construction. Selection of VHHs recognizing BDE-47 was achieved by panning under carefully modified conditions.
Specificity: The antibody is specific for BDE-47 and it does not cross react with BDE-153, BDE-154, BDE-183, triclocarbon and TBBPA. BDE-47 is a polybrominated diphenyl ether which has been used as flame retardant.
Application Notes: The specificity of the VHH fragment was confirmed by cELISA analysis. The VHH fragment showed increased thermal stability compared to polyclonal antibodies. The VHH antibody was immobilized onto an Au electrode and used for impedance biosensing, showing a detection limit for BDE-47 of approximately 0.79 μg/L (Bever et al., 2014; PMID: 25005746).
