Immunogen: The original antibody was generated by immunizing BALB/c mice with a mixture of transfected HEK 293 cells expressing human and mouse β-DAG.
Specificity: This antibody recognizes human and mouse beta dystroglycan (β-DAG). Beta-dystroglycan is a transmembrane protein that plays an important role in connecting the extracellular matrix to the cytoskeleton. It plays an important role in linking dystrophin to the laminin-binding alpha-dystroglycan. It acts as a cell adhesion receptor in both muscle and non-muscle tissues. It also functions in cell adhesion-mediated signaling and implicated in cell polarity.
Application Notes: The binding specificity of this antibody to human and mouse beta dystroglycan expressed on the surface of CHO cells was confirmed using ELISA and flow cytometry. The binding affinity of the original IgG antibody to human beta dystroglycan was determined to be 941 pM. This antibody was used in combination with anti-Laminin antibody PL60B3 for the generation of a bispecific antibody (CN113651891).

Flow cytometry using anti-Beta Dystroglycan antibody P38G1 (Ab03279). MCF7 cells were stained with the anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of P38G1 (Ab03279-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.
Western blot using anti-Beta Dystroglycan antibody P38G1 (Ab03279). Human skeletal muscle tissue lysate (35µg protein in RIPA buffer) was resolved on an SDS-PAGE gel, and blots were probed with the chimeric rabbit version of P38G1 (Ab03279-23.0) at 0.1µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1 hour was used, and protein was detected by chemiluminescence.
