Recombinant monoclonal antibody to CD147. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma OX-47.

Immunogen: Mice immunized with rat T blasts from a mixed lymphocyte reaction between purified CD4+ T cells and irradiated spleen cells.
Specificity: This antibody is specific for rat CD147, which has a broad distribution; OX-47 labels T and B cell blasts, peritoneal macrophages, the endothelium of all except large vessels and kidney tubules, but does not label TDL and thymocytes.
Application Notes: This antibody binds mouse CD147, and has been used in FC and IHC-fr analysis to characterise CD147 and detect the cell types on which it is expressed, indicating that CD147 is a lymphocyte activation marker (Paterson, 1987). This antibody failed to give immunoprecipitates from radiolabelled activated T cells.

Flow-cytometry using anti-CD147 (Basigin) antibody OX-47 (Ab00573) Staining of rat (Rattus norvegicus) leukocytes with either an isotype control (A) or the rabbit-chimeric version of OX-47 (B, Ab00573-23.0) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using an Alexa Fluor ® 488 conjugated donkey anti-rabbit antibody (ab150073) and cells analysed on a FACSCanto flow-cytometer.
Immunofluorescence staining of fixed rat splenocyte cells with anti-CD147 (Basigin) antibody OX-47 (Ab00573) Immunofluorescence analysis of paraformaldehyde fixed rat (Rattus norvegicus) splenocytes on Shi-fix™ coverslips, permeabilized with 0.15% Triton stained with the chimeric rabbit version of OX-47 (Ab00573-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (1 µg/ml), showing membrane staining of a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00573-23.0, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody (Ab102-23.0) followed by Alexa Fluor® 488 secondary antibody.
Western Blot using anti-CD147 (Basigin) antibody OX-47 (Ab00573) Rat thymus tissue lysate (35µg protein in RIPA buffer) was resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of OX-47 (Ab00573-23.0) at 0.1 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected band size for CD147 is 42.4kDa. Ab00573-23.0 successfully detected CD147 in rat thymus tissue lysate.
