Recombinant monoclonal antibody to CD204. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma 2F8 (recombinant version).
UniProt Accession Number of Target Protein: P30204
Alternative Name(s) of Target: Macrophage scavenger receptor; Murine scavenger receptor class A; Macrophage scavenger receptor types I and II; Macrophage acetylated LDL receptor I and II; Scavenger receptor type A; SR-A
Immunogen: Rats immunised with Raw 264 cell line.
Specificity: This antibody specifically recognizes CD204, also known as the the murine scavenger receptor class A (SRA), type I and II. which is expressed by tissue macrophages and functions both as an endocytic receptor for lipoproteins and as an adhesion receptor for macrophages binding to ligand rich tissues e.g. atherosclerotic lesions.
Antibody first published in:
Fraser I, Hughes D, Gordon S. Divalent cation-independent macrophage adhesion inhibited by monoclonal antibody to murine scavenger receptor. This antibody has been shown to be endocytic. Nature. 1993 Jul 22;364(6435):343-6. PMID:8332192
Note on publication:
Describes the making of 2F8 monoclonal antibody and used for characterization of macrophage adhesion.
Immunofluorescence staining of RAW264.7 cells with anti-CD204 (Ab00163) 2F8 (recombinant version) Immunofluorescence analysis of paraformaldehyde fixed RAW264.7 cells stained with the chimeric rabbit IgG version of 2F8 (recombinant version) (Ab00163-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (2 µg/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00163-23.0, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody (Ab00178-23.0) followed by staining with Alexa Fluor® 488 secondary antibody.
Western Blot using anti-CD204 antibody 2F8 (recombinant version) (Ab00163). RAW264.7 cell lysate of (35µg protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of 2F8 (recombinant version) (Ab00163-23.0) 0.3 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Flow cytometry using the Anti-CD204 antibody 2F8 (recombinant version) (Ab00163). Paraformaldehyde fixed RAW264.7 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of 2F8 (recombinant version) (Ab00163-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.
