Immunogen: The original antibody was generated by immunizing mice with rat lymphocytes or rat thymocyte glycoproteins.
Specificity: This antibody binds rat CD4 cell surface antigen. CD4 is a transmembrane glycoprotein expressed on T helper cells, thymocytes, monocytes and macrophages. It plays an important role in the response of T helper cells to antigen. This antibody binds an identical or adjacent epitope to that bound by W3/25 antibody, and competes with mouse anti-rat CD4 antibody W3/25 for binding rat CD4.
Application Notes: This antibody can be used to identify CD4 antigen present on T cells, thymocytes, monocytes etc. using flow cytometry. This antibody was also able to immunoprecipitate detergent extracts of iodine 125 labeled thymocytes (PMID: 3159821). This antibody can be used for the in vitro depletion of T helper cells from slice cultures in studies looking for myelin loss in multiple sclerosis, the observed disruption being abrogated in these experimental conditions by T-cell depletion (PMID: 25500111). This antibody was tested for its ability to prolong the survival of heterotopic vascularized rat heart allografts transplanted across major histocompatibility barriers. Nine of 12 treated rats have maintained heterotopic abdominal heart allografts for greater than 175 days (PMID: 1976282). This antibody was used in a study to demonstrate that monoclonal antibody-induced depletion of CD4 helper/inducer T lymphocytes before transplantation of a fully mismatched heart allograft allows permanent engraftment in rats without further immunosuppression (PMID: 1977898).

Flow cytometry using anti-CD51 antibody NKI-M9 (Ab02884). HeLa cells were stained with the anti-unknown specificity antibody (Ab00178-23.0; isotype control, black line) or the rabbit IgG version of NKI-M9 (Ab02884-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000 and the cells were analyzed using a FACSCanto flow-cytometer.
