Immunogen: OX-78 was prepared by immunising rats against mouse CD48.
Specificity: OX-78 binds specifically to CD48 targets and labels mouse thymocytes, B and T cells and macrophages. CD48 is a CD2 ligand and may be involved in the facilitation of interactions between activated lymphocytes. This protein is also likely to be involved in T cell activation.
Application Notes: This antibody binds CD48, and has been used in immunoprecipitation and blocking studies to identify and characterise CD48, the ligand for murine CD2 (Kato, 1992). This antibody is able to block the binding of a chimeric molecule comprised of the extracellular domain of mouse CD2 and human IgG1 Fc to T cell lines at the ligand site.

Immunofluorescence staining of fixed mouse thymocytes with anti-CD48 antibody OX-78 (Ab00574) Immunofluorescence analysis of paraformaldehyde fixed mouse (Mus musculus) thymocytes on Shi-fix™ coverslips, permeabilized with 0.15% Triton stained with the chimeric rabbit version of OX-78 (Ab00574-23.0) at 10 µg/ml for 1h followed by Alexa Fluor® 488 secondary antibody (1 µg/ml), showing membrane staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom Ab00574-23.0, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody (Ab102-23.0) followed by Alexa Fluor® 488 secondary antibody.
Western Blot using anti-CD48 antibody OX-78 (Ab00574) Mouse spleen (A), thymus (B) and lymph node (C) tissue lysate samples (35µg protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of OX-78 (Ab00574-23.0) at 0.0001 µg/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for CD48 is ~37-56kDa and it is heavily glycosylated, as well as lipidated (UniProt ID: P18181). Ab00574-23.0 successfully detected CD48 in mouse spleen, thymus and lymph node tissue lysates.
