Recombinant monoclonal antibody to E protein. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma 13D6.

Immunogen: The original antibody was generated by immunizing BALB/c mice with purified E protein.
Specificity: The antibody binds to domain III of glycoprotein E of TBEV. Surface glycoprotein E has a major role in flavivirus biology, inducing protective immune responses in vivo, and participating in cell receptor binding and erythrocyte agglutination.
Application Notes: The specificity of the IgG1 format was confirmed by ELISA analysis (Tsekhanovskaya et al. 1993; PMID: 7505512). The affinity constants of the chimeric, scFv and parental antibody were evaluated by ELISA. The affinity constants of the chimeric and scFv were respectively, two and three times higher than the ones of the parental antibody. The parental, chimeric and scFv version exhibited binding characteristics similar to parental antibody. The chimeric and scFv versions were able to neutralize TBEV infectivity in vitro. The parental antibody showed an IC50 titer of 11.5 𝜇g/mL in FRNT against TBEV strain 205 and of 2.9 𝜇g/mL in PRNT against TBEV strain Softjin. The chimeric showed an IC50 titer of 4.5 𝜇g/mL in FRNT against TBEV strain 205 and of 1.9 𝜇g/mL in PRNT against TBEV strain Softjin. ScFv fragment showed an IC50 titer of 16.7 𝜇g/mL in FRNT against TBEV strain 205 and of 11.2 𝜇g/mL in PRNT against TBEV strain Softjin. These results suggest this antibody has potential for therapeutic uses (Levanov et al. 2010, PMID: 20538092). This antibody was used for immunohistochemistry against the E protein of TBEV strain Sofjin on monkey tissue from the spinal cord, frontal cortex, thalamus and cerebellum (Maximova et al., 2008; PMID: 18353947). Immunofluorescence was performed on Vero E6 cells using this antibody (Levanov et al. 2014, PMID: 24371235).
