Recombinant monoclonal antibody to F. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the phage display antibody DS7.
UniProt Accession Number of Target Protein: Q6WB98
Alternative Name(s) of Target: hMPV; Fusion glycoprotein F0; Protein F; DSλ7
Immunogen: Phage antibody libraries were prepared from human donor tissues. The original antibody was selected against recombinant hMPV F protein with multiple rounds of panning.
Specificity: The antibody is specific for HMPV. It cross-reacts with lineage A1, A2, B1.
Application Notes: The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody could specifically bind to hMPV-infected cell monolayers as shown by immunofluorescent assays. The Fab fragment neutralized the parent strain of hMPV with a 60% plaque reduction activity of 1.1 ug/ml and bound to hMPV F with an affinity of 9.8 10^-10 M, as measured by surface plasmon resonance. The in vivo activity of the Fab fragment was tested on groups of cotton rats infected with hMPV. Nasal turbinates and lungs were harvested on day 4 postinfection and virus titers determined. Animals treated with the Fab fragment exhibited a >1,500-fold reduction in viral titer in the lungs, with a 4-fold reduction in the nasal tissues (Williams et al. 2007; PMID:17522220).
