Immunogen: The original antibody was generated by immunizing BALB/cJ mice intradermally with Francisella tularensis holarctica strain LVS (live vaccine strain) and later on a booster immunization with an outer membrane-enriched LVS preparation of F. tularensis.
Specificity: This antibody binds the non-reducing end of the O-antigen (OAg) of the gram-negative bacterium Francisella tularensis (Ft). The antibody N62 binds mainly to the terminal Qui4NFm sugar residue at the nonreducing end of OAg.
Application Notes: This antibody binds the same epitope as clone FB11 and has a higher antigen-binding bivalent avidity than internally binding antiOAg mAb. This antibody is capable of detecting F. tularensis SchuS4 strain in an ELISA. It is also capable of agglutinating F. tularensis SchuS4 and live vaccine strain. This antibody also binds strongly to both long and short lipopolysaccharide chains in western blot assays. In efficacy studies with mice infected intranasally with the highly virulent F. tularensis strain SchuS4, N62 prolonged survival and reduced blood bacterial burden (PMID: 23844703)
