Recombinant monoclonal antibody to gp120. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the phage display antibody Y498.

Immunogen: A phage display Fab library was constructed using the PBMC samples of a CRF07_BC-infected subject. The antibody was selected by panning the library with a CRF07_BC (CN54)-derived rgp120 antigen.
Specificity: The antibody is specific for gp120. The conformational epitope of contains three antigenic sites on gp120, including the CD4 binding loop in C3, the β23 in C4 and the β24-α5 in C5.
Application Notes: The Fab fragment neutralized the R5 isolate SF162 and the X4 isolate HXB2 at a dose-dependent manner. The specificity of the Fab and IgG1 format of the antibody was confirmed by ELISA analysis. The binding affinity of the IgG1 antibody was measured by SPR assay, the antibody bound to CN54 rgp120 with a Kd value of 0.46 nM and bound to JRFL rgp120 with a Kd value at 0.86 nM. The antibody cross-reacted with diverse Env antigens and neutralized 30% of 70 tested HIV-1 isolates. The antibody could efficiently inhibit the binding of biotinylated sCD4 to rgp120 at a dose-dependent manner in competition ELISA (Sun et al., 2017; PMID :28968970).
