Immunogen: The original antibody was isolated using phage display from the IgM+ memory B cells of a healthy individual vaccinated against influenza.
Specificity: This antibody binds hemaglutinin of most group 1 influenza viruses. CR6261 recognizes a highly conserved helical region in the membrane-proximal stem of HA1/HA2.
Application Notes: The antibody neutralizes the virus by blocking conformational rearrangements associated with membrane fusion. CR6261 neutralizes multiple influenza subtypes, including H1, H2, H5, H6, H8, and H9, and protects mice from lethal challenge with H1N1 and H5N1 viruses when administered up to five days post-infection (PMID: 19251591). It is reported that Oseltamivir/Zanamivir may congruously improve the therapeutic efficacy of CR6261 (PMID: 22693576).

ELISA using CR6261 (Ab02199) and Influenza A HA [A/Hong Kong/483/97 (H5N1)]. Binding curves of the human IgG chimeric version of the Anti-HAHA antibody CR6261 (Ab02199-10.0; blue line) and isotype control (Ab00102-10.0, anti-fluorescein; orange line) to an ELISA plate coated with Influenza A HA [A/Hong Kong/483/97 (H5N1)] at a concentration of 2.5 ug/ml. A 3-fold serial dilution from 3000 to 0.0169 ng/ml was performed using Ab02199 antibody. For signal detection, a 1:4000 dilution of a HRP-conjugated anti-human IgG antibody was used.
