Recombinant monoclonal antibody to HA. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the phage display antibody Mab 3.1.

Immunogen: A Fab phage display library was prepared from mature B cells of a healthy donor and the original antibody was isolated by panning against HA from A/Japan/305/1957 (H2N2) virus.
Specificity: The antibody recognizes the conserved heterosubtypic epitope in the stem of influenza A virus hemagglutinin. The epitope consists of residues from the N- and C-terminal regions of HA1 (38, 40 to 42, 289 to 293, and 318) and the N-terminal portion of HA2 (18 to 21, 38, 41, 42, 45, 49, 52, 53, 56), including helix A. The antibody neutralizes influenza viruses from the H1a clade but has little neutralizing activity against the H1b clade. The antibody binds HA proteins from A/Puerto Rico/8/1934(H1N1), and A/Japan/305/1957(H2N2), A/flat-faced bat/Peru/033/2010(H18N1) virus, A/duck/Alberta/345/1976(H1N1), A/USSR/90/1977(H1N1), A/Beijing/262/1995(H1N1), A/Solomon Islands/3/2006(H1N1), A/Adachi/2/1957(H2N2), A/Vietnam/1203/2004(H5N1), and A/turkey/Massachusetts/3740/1965(H6N2) viruses. It fails to bind to A/Victoria/3/1975(H3N2) and A/fowlplague/Bratislava/1979(H7N7), A/little yellow-shouldered bat/Guatemala/164/2009(H17N10) virus,A/duck/Alberta/60/1976(H12N5), A/gull/Maryland/704/1977(H13N6), A/black-headed gull/Sweden/4/1999(H16N3), A/duck/Ukraine/1/1963(H3N8), A/Hong Kong/1/1968(H3N2), A/duck/Czechoslovakia/1956(H4N6), A/Netherlands/219/2003(H7N7), A/chicken/Germany/N/1949(H10N7), A/mallard/Astrakhan/263/1982(H14N5), or A/shearwater/W. Australia/2576/1979(H15N9) viruses.
Application Notes: In ELISAs, the Fab fragment was found to bind to recombinant HA protein. The Fab fragment neutralized viruses of the H1, H2, H5, and H6 subtypes at half-maximal inhibitory concentrations in the ug/ml range. Passive immunization with the Fab fragment protected mice from a lethal challenge with A/Puerto Rico/8/1934(H1N1) virus. The crystal structure of the antibody in complex with HA from a pandemic H1N1 influenza virus was determined.
