Immunogen: A combinatorial antibody library of γ1-heavy and κ-light chain genes was constructed from bone marrow cells of a chimpanzee experimentally infected with hepatitis B virus (HBV). The antibody library was displayed on the surface of bacteriophage particles. The original antibody was selected by panning on hepatitis B surface antigen (HBsAg).

Specificity: The original antibody is specific for HBV.

Application Notes: The affinities of both the Fab and IgG fragments of the antibody to HBsAg were determined by competition inhibition ELISA. The concentration of free HBsAg required to inhibit antibody binding by 50% is equivalent to the equilibrium dissociation constant (Kd). The Kd value the Fab fragment was 1.9×10^−6 M, and 2.5×10^−8M for the IgG. The ability of the Fab fragment to neutralize HBV in a primary hepatocyte cell culture system was determined. The Fab fragment could neutralize HBV (highest concentration tested= 10 μg/ ml). The levels of HBsAg in cell supernatants were reduced by >90% compared the control, and levels of HBV DNA in the cell lysates were diminished.
