Recombinant monoclonal antibody to HCMV gH. Manufactured using AbAb’s Recombinant Platform with variable regions (i.e. specificity) from the hybridoma HCMV16.

Immunogen: The original antibody was generated by immunization of mice with purified HCMV strain AD169.
Specificity: This antibody binds gpUL75 (gH) of the human cytomegalovirus, which is an 86-kDa glycoprotein. The heterodimer glycoprotein H-glycoprotein L is required for the fusion of viral and plasma membranes leading to virus entry into the host cell. It is though to bind a conformational epitope on the gH, and its bind is destroyed if the protein is denatured.
Application Notes: This antibody successfully precipitates glycoprotein H from both HCMV infected cell lysates and HCMV-gH-VAC-infected cell lysates. Cellular localization of HCMV-gH studied by immunofluorescence on recombinant vaccinia virus and HCMV-infected cells using HCMV-16. HCMV-16 strongly neutralizes HCMV infectivity in vitro (PMID: 2831402). The reshaped humanized antibody recognized cells infected with a variety of virus strains and neutralized clinical isolates of CMV as efficiently as laboratory strains in a conventional plaque reduction assay (PMID: 9207350).

Describes the expression of glycoprotein H and sequence homology between HCMV gH and Epstein-Barr Virus BXLF2 Product, Varicella-Zoster Virus gpIII, and Herpes Simplex Virus Type 1 Glycoprotein H. Mab HCMV-16 was used for the identification of gH.
ELISA using HCMV16 (Ab02066) and Cytomegalovirus gH Pentamer Complex. Binding curves of the rabbit IgG chimeric version of the Anti-HCMV gHHA antibody HCMV16 (Ab02066-23.0; green line) and isotype control (Ab00102-23.0, anti-fluorescein; orange line) to an ELISA plate coated with Cytomegalovirus gH Pentamer Complex at a concentration of 2.5 ug/ml. A 3-fold serial dilution from 3000 to 0.0169 ng/ml was performed using Ab02066 antibody. For signal detection, a 1:4000 dilution of a HRP-conjugated anti-rabbit IgG antibody was used.
